Wound Healing Activity of Bone Marrow Stromal Cells

ABSTRACT
Bone marrow stroma is the source of mesenchymal stem cells. The bone marrow stromal stem cells (BSCs) secrete collagen and several cytokines essential for wound healing. However, it is unclear as to what extent these materials are produced and it has not yet been studied whether they are superior to fibroblasts (which have been used conventionally) in promoting the wound healing process. It has been hypothesized that BSCs and fibroblasts would differ in their ability to promote wound healing, and furthermore that BSCs should have greater activity than fibroblasts. If so, then the current cell therapy method using fibroblasts to stimulate the healing of chronic wounds could be replaced by a therapy which uses BSCs and has a superior effect. The purpose of this review is to present the author's studies comparing the wound healing activity of BSCs with that of fibroblast. Cultured human BSCs and dermal fibroblasts taken from same patients were tested. In vitro study was focused on cell proliferation, synthesis of collagen, and production of three growth factors - basic fibroblast growth factor (bFGF), vascular endothelial growth factor (VEGF), and transforming growth factor beta (TGF-Ղ). In vivo study was performed to compare collagen synthesis, epithelization, and angiogenic activity of BSCs with those of fibroblasts. The results suggest that the potential of BSCs in wound healing acceleration may be superior to that of fibroblasts and that BSCs may be possibly used as a replacement for fibroblasts, which are currently being used for wound healing.
